My favorite is Castlevania Bloodlines for the Sega Genesis. That is the one I play the most because I am a Genesis guy and my Genesis is always hooked up. I also have Simons Quest but I am not a big fan of that one, the music in it is good though.

Super Castlevania 4, Aria of Sorrow, Rondo of Blood, and Symphony of the Night were amazing in my opinion. I especially love Symphony of the Night, just pure genius in the music graphics gameplay just everything went together for an awesome 2D experience.

I really enjoy the original CastleVania for the NES. Some people are put off by the difficulty but I am a person who enjoys a hard game, when it isn't full of cheapness like most fighting games. Even making a single jump feels progressive and rewarding and the soundtrack compliments the gameplay well. 
Some of the MetroidVania games are good like Sympathy of the Night, I got the first Lord of Shadows game and didn't like it and recently tried a demo of the second on steam and still felt negative towards them.

I did enjoy playing it on my Korean handheld system, CAANOO.

Even though the rom I download made the game's soundtrack sped up times 2x. Sounded funny when it came to certain theme's.

If I would pick the ones from the subjects at hand, easily I would pick Dracula's Curse. It was a full fledged save from the failure of the 2nd Game of Simon's Quest, with an RPG-esque type of game. Instead, back to the basics that got the franchise famous in the first place.
